I have been from one end to the other on foot, motorcycles, and cars and can't think of too many places where people would have homesteads, farms, etc.
That project was an economic boom for one of the most depressed areas in America, Appalachia.

I have and have not found anything. As I stated earlier, I am curious, mostly because my own experience is that the Skyline Drive doesn't seem to traverse the landscape that would be suitable for homesteads, farms, etc. In any case, building that roadway brought lots of benefits to the people of the area. Certainly well worth displacing a small number of people if it did.
