I don't know which is the most powerful. The several that are most often cited -- the AIPAC, NRA, Big-Pharma, Finance, Agribusiness, Mining, Oil and Gas, Defence and Tech -- are without a doubt quite vocal, but I'd wager that it's the lobby that is nearly never cited that is the most powerful:
The Chamber of Commerce. (See also:

That shouldn't surprise anyone. What are the factors of production? Land, labor, capital and entrepreneurship. Quite simply, in the story of any economic boom, while labor and capital variously enjoy their days in the sun, so to speak, land and entrepreneurship never fall from favor. Perhaps land will suffer when we figure out how to build in midair, thus reducing land's enduring value, but until then.....After all, even at Hiroshima and Nagasaki, the land didn't go anywhere; it just needed some time to be habitable and usable again. Entrepreneurship is what will use it, and entrepreneurship is all the Chamber is about.
I would venture to say there's not a single policy or regulatory matter on which the Chamber doesn't have a very deeply knowledgeable expert of the first caliber and that the Chamber doesn't vociferously lobby for. Unlike many other lobbies, the Chamber doesn't shy away from hauling the government into court, regardless of the administration. The Chamber is doggedly about itself and its members. Period.
The Chamber was essentially created by the government itself and it's got special tax exempt status that keeps hidden nearly all its sources of funding. It's also the one lobbying entity that routinely and consistently supports both Democrats and Republicans. Though as an industry, Oil and Gas outspend the Chamber, as a single entity, it's the Chamber.
Anyway, that's my opinion on the matter of what lobby is most powerful. Sorry for the off topic digression...carry on.....
