Here Sotomayor forces the attorney arguing that boys should not be allowed on girls teams,* to give an answer based on a hypothetical in which there is no biological difference between men and women.
Giving Sotomayor more credit that maybe I should, I think that question indicates that she knows that of course there is a difference. She may be a surprise vote for the correct side. She may have played girls sports herself once and would understand how absurd it is to even consider allowing boys on the girls teams.

*Really, he was arguing that Title IX does not forbid schools from keeping boys off the girls teams.
