Jesus said before he died on the cross that he commanded his spirit to God. But we also know that He spent the next day in hell. So if his spirit was in the hand of God, then it was not his spirit but his soul that spent that Saturday in hell. So what is the difference between soul and spirit?

Sometimes there used interchangeably.
A lot of times it means the body and the spirit united.
But it can mean a dead person's spirit also.
It's really not that hard to decipher when you look at the context
Don't make it harder than it is
 
The difference between the spirit and the the soul is that the soul is the combination of both the body and the spirit and the spirit is that spirit which existed prior to the body. Before coming to this earth, we all existed in the presence of God as his spirit children.

Hebrews 12:9
9 Furthermore we have had fathers of our flesh which corrected us, and we gave them reverence: shall we not much rather be in subjection unto the Father of spirits, and live?

Ecclesiastes 12:7
7 Then shall the dust return to the earth as it was: and the spirit shall return unto God who gave it.

That's actually a good question.

To humans, we have a soul as our permanent identity. As an analogy, if murderer is dead, is it a fair trial if we clone a person with the same exact DNA to stand the trial? Similarly, if God resurrected a person to stand trial, then no one except for God alone knows whether he's the same person or a clone. A soul is to make a continuous witnessing possible, such that the angels or chosen saints (i.e., entities other than God) can witness who this person actually is.

A spirit is needed to carry what is necessary for a human to live his life on earth. It is needed because a human's eternal life is staged. On earth we have different kind of earthly affections, to our parents, to our children etc. In heaven however our affection is limited to brothers and sisters. That's why a spirit is needed to carry the stage (basically earthly stage and heavenly stage) specific elements. In hell then, you don't need a spirit, it means you have no parent, no kids, no brothers and sisters or any relatives in terms of emotions. They are more or less like your ex girlfriends. You may still remember them, but your affection now is on your wife, your affection to your EXs is gone.

Humans have a soul. God is Spirit. The Adversary is spirit. We need to be able to differentiate between the two, not allowing the Adversary to come into our soul.

A clear misunderstanding of scripture. Read (1 Peter 3:18-20). "For Christ also suffered once for our sins, the just for the unjust, that He might bring us to God, being put to death in the flesh but made alive BY THE SPIRIT, BY WHOM (the spirit) He went and preached to the spirits in prison, who formerly were disobedient, when once the longsuffering waited in the days of Noah, while the ark was being prepared, in which a few, that is, 8 souls, were saved through water."

First you must consider who this passage of scripture is talking about.......the subject of the passage is HE (Jesus)....discussed in v.18. Second, what did Christ do? He preached or proclaimed to the spirits in prison, Who were these SPIRITS He preached to.....explained in v. 20 The spirits in prison were the disobedient people who lived on earth during the days that Noah was constructing the ARK.

Next we must ask HOW DID JESUS DO THIS PREACHING? The passage identifies this process with "which" or "by whom" at the very beginning of the passage and last mentioned in v. 18. Answer is professed in the passage. We were made alive through the same means that Christ preached to the spirits in prison........through the SPIRIT....by this means Jesus preached to the imprisoned spirits. Take care to note: This passage does not declare that Jesus preached directly to the spirits in prison. The proclamation was accomplished THROUGH THE SPIRIT.....not the soul. Jesus preached IN SPIRIT....a spirit has no bones, blood, flesh....etc.

There is a difference between HELL and HADES........one is the place where the dead go to await the judgement, i.e., the SPIRIT given by God produces or animates life........ the other is a place of everlasting torment.
